响应面法优化艾蒿水提物的提取工艺及其抗氧化活性分析

摘要:
为优化艾蒿水提物的提取工艺,并探究其抗氧化活性,在单因素试验的基础上,以料液比、水浴时间和温度为自变量,艾蒿水提物的提取率为响应值,通过响应面分析得到艾蒿水提物的最佳提取工艺;通过测定铁还原力、1,1-二苯基-2-三硝基苯肼自由基(DPPH)和羟自由基(·OH)的清除能力,评价艾蒿水提物的抗氧化活性。结果表明,艾蒿水提物的最佳提取工艺为:1∶25(艾蒿质量与水的体积比),84 ℃水浴10 h,所得艾蒿水提物的提取率为18.05%。不同浓度提取液的铁还原力与样品浓度呈正相关,提取液清除DPPH自由基和·OH自由基的半清除率IC50分别为0.102和0.311 mg/mL。综上,本优化工艺不仅提取率较高,而且所得水提物具有较强的抗氧化活性。

Optimizing the aqueous extraction process of Artemisia argyi by response surface methodology and its antioxidant activity analysis

Abstract:
The study was conducted to optimize the extraction conditions for Artemisia argyi aqueous extraction(AAE)and investigate the antioxidant activity of AAE. Based on the single-factor test, response surface methodology was used to optimize the extraction conditions of AAE. The extraction rate of AAE was considered as the response value, and the solid-liquid ratio, time and temperature for water as a solvent were used as independent variables. The antioxidant activity of AAE was evaluated by determining iron reduction capacity, scavenging ability of DPPH and hydroxyl free radical. The results showed that: The optimal extraction conditions with water as extraction solvent were as follows: solid-liquid ratio 1∶25(the ratio of Artemisia argyi mass to water volume), the extraction temperature 84 ℃ and time 10 h. The extraction rate of AAE was 18. 05%. There was a positive correlation between AAE concentration and iron reduction capacity, and the IC50 of DPPH and hydroxyl free radical was 0. 102 and 0. 311 mg/mL, respectively. In conclusion, the results suggested that this optimal condition could be used to get higher extraction rate of AAE with strong antioxidant potential in vitro.
