红花蝴蝶兰新品种选育与遗传多样性分析

摘要:
为提供红花蝴蝶兰新品种选育与育种亲本选择的科学依据,采用形态学性状评价和SRAP分子标记,研究来自广东省农业科学院环境园艺研究所自主培育的同一杂交群体的7个蝴蝶兰新品种、3个株系及其父母本‘聚宝红玫瑰’蝴蝶兰和‘红龙’蝴蝶兰的形态特征和遗传多样性差异。首先,形态学性状评价结果表明,子代具明显的倾母本遗传,遗传变异程度较低,基本不具备超亲遗传;其次,对12份蝴蝶兰进行SRAP分析,筛选出带型稳定、多态性较好的24对引物,共扩增出172条谱带,平均每对引物扩增7.17条,最多10条,最少4条,其中多态性条带99条,多态性比率58.14%,根据UPGMA聚类图,在遗传距离0.778处,可把亲本和后代分成4类,这与根据形态特征的聚类结果具有较高程度的吻合和较小的差异。综合形态学性状评价结果和分子标记结果可发现,由于亲本相似性程度较高,导致子代的遗传分化较小,不利于优良新品种的选育。

Breeding and genetic diversity of new red Phalaenopsis varieties

Abstract:
Morphological character analysis and SRAP molecular marker analysis were used to study the genetic diversity of seven new Phalaenopsis varieties and three lines,which were selected and bred from the progenies of Phalaenopsis ‘Jubao Red Rose’ and Phalaenopsis ‘Ben Yu Star’ by Environmental Horticulture Research Institute of Guangdong Academy of Agricultural Sciences.The results of morphological character analysis showed that the offspring had obvious diploid maternal inheritance,low degree of genetic variation and almost no superparental inheritance.On the other hand,a total of 24 primer pairs with stable bands and good polymorphism were screened.A total of 172 bands were amplified with an average of 7.17 fragments,up to 10,and at least 4 per pair of primer among which 99 (58.14%) were polymorphic bands.Based on the amplification results of SRAP markers,the genetic similarity coefficients of GS were calculated by using NTSYS 2.10e software,and a UPGMA clustering map was established.At the genetic distance of 0.778,the parents and offspring could be divided into 4 subgroups,which were closely in line according to the morphological characteristics.The results of morphological character analysis and molecular markers showed that the genetic differentiation of offspring was small due to the high degree of parental similarity,which was not conducive to the breeding of new varieties.
